Inhibitory effect of the methanolic leaf extract of Mormodical charantia on Ca2+-induced liver mitochondrial permeability transition pore opening in normal rats.

Abstract

The involvement of mitochondria in the orchestration of apoptosis via the intrinsic pathway has been well established and opening of the mitochondrial permeability transition pore (MPTP), a non-specific pore in the inner mitochondrial membrane, pinpointed as a critical stage of this death. The inhibitory effect of varying concentrations (0.2, 0.4, 0.6, 0.8 and 1.0mg/ml) of the methanol leaf extract of Momordica charantia on Ca2+-induced MPTP opening in liver mitochondria isolated from normal rats was studied. Mitochondrial swelling induced by 300?M CaCl2/mg mitochondrial protein was quantitatively measured at 540 nm based on a modification of the procedure described by Lapidus and Sokolove (1993). Results obtained showed significant (P<0 xss=removed xss=removed> 0.8mg/ml > 0.6mg/ml > 0.4mg/ml > 0.2mg/ml (59%>52%>48%>45%>0%) by extract alone and (76%>74%>73%>63%>37.2%) by combined extract and spermine. The degrees of inhibition from 0.6mg/ml upward suggest that the methanol extract is an MPTP antagonist and that this inhibition could be the mechanism by which M. charantia inhibits pathological cell loss in diseases arising from excessive apoptosis. Potentiated inhibition of MPTP in the presence of spermine, a specific inhibitor of the pore confirms that the mitochondrial swelling observed in this study is via MPTP opening.
